I recently bought a 320GB USB HDD for backup purposes -- and it works fantastically well! After backing everything up to it, and reformatting both my laptop and desktop, I now occasionally need to grab a file or two off it that I didn't think I needed to restore at the time.

My question -- is it ok to power the drive on and off as needed? Or should I turn it on and leave it on for the day? Or 24/7? I just don't want to destroy my (now invaluable!) backup device ...

You should be fine turning it on and off as needed.
Hard drives these days are made to do this anyway to save power when they are not being used. Don't think a USB one would do this but in a PC or laptop they do.

Just make sure you stop the Removal Drive before turning it off just in case it is still being read or written to.
